CTLA4 polymorphisms and COPD.

چکیده
We read with interest the article by ZHU et al. [1] describing the association of two single nucleotide polymorphisms (SNPs; rs231775 and rs3087243) within the CTLA4 gene and chronic bronchitis. Both of these polymorphisms may be risk factors for autoimmunity [2], and are often referred to as A49G and CT60, respectively. As alluded to briefly by the authors in their discussion, there has been recent interest in a possible role for autoimmunity in chronic obstructive pulmonary disease (COPD), hence shared genetic susceptibility with other autoimmune diseases would support this hypothesis. Although the authors report their association in two independent cohorts, general acceptance of the importance of this locus would require further validation in other COPD populations. We would like to add support to their results, by reporting association of rs231775 in an a1-antitrypsin deficient (a1-ATD) cohort, which also tended to associate with the same feature in the 1958 birth cohort.

CTLA4 gene polymorphisms are associated with chronic bronchitis.
Chronic obstructive pulmonary disease (COPD) is characterised by chronic and progressive dyspnoea, cough and sputum production. T-lymphocytes may play a key role in the pathogenesis of COPD and chronic bronchitis. Cytotoxic T-lymphocyte antigen (CTLA) 4 is a potential candidate gene because it modulates T-cell activation. Genetic association between nine CTLA4 single nucleotide polymorphisms (S...
متن کاملP-212: Association between Polymorphisms of CTLA-4 Gene and Unexplained Recurrent Spontaneous Abortion in An Iranian Population
Background: Unexplained recurrent spontaneous abortion (URSA) has been suggested being associated with the failure of fetal-maternal immunologic tolerance in which the regulatory T lymphocytes (Tregs) play a crucial role. Cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) is expressed transiently on activated T cells and constitutively on Tregs. CTLA-4 interacts with the B7 cell surface molec...
متن کاملCTLA4 Gene Variants in Autoimmunity and Cancer: a Comparative Review
Gene association studies are less appealing in cancer compared to autoimmune diseases. Complexity, heterogeneity, variation in histological types, age at onset, short survival, and acute versus chronic conditions are cancer related factors which are different from an organ specific autoimmune disease, such as Grave’s disease, on which a large body of multicentre data is accumulated. For years t...
متن کاملTechnical note: linkage disequilibrium and disease-associated CTLA4 gene polymorphisms.
CTLA4 and CD28 are important regulators of T lymphocyte activation. Gene region 2q33 carrying genes for both CTLA4 and CD28 has been shown to be linked to many autoimmune diseases. Disease associations with particular CTLA4 gene polymorphisms have been reported. Recently, first lines of evidence emerged for functional effects of CTLA4 gene polymorphisms. Two independent studies reported a reduc...
متن کاملAssociation of Cytotoxic T Lymphocyte Antigen-4 Gene Polymorphisms and HLA Class II Alleles with the Development of Type 1 Diabetes in Korean Children and Adolescents
We studied the association of cytotoxic T lymphocyte antigen-4 gene (CTLA4) polymorphisms with the development of type 1 diabetes (T1D) in Korean children and adolescents. A total of 176 Korean subjects (92 females and 84 males) with childhood-onset T1D were studied. The A/G polymorphism at position 49 in CTLA4 exon 1 and the C/T polymorphism at position -318 in the CTLA4 promoter were analyzed...
